    Using biomarker signature patterns for an mRNA molecular diagnostic of mouse embryonic stem cell differentiation state

    BACKGROUND: The pluripotency and self-renewal capabilities, which define the 'stemness' state, of mouse embryonic stem (ES) cells, are usually investigated by functional assays or quantitative measurements of the expression levels of known ES cell markers. Strong correlations between these expression levels and functional assays, particularly at the early stage of cell differentiation, have usually not been observed. An effective molecular diagnostic to properly identify the differentiation state of mouse ES cells, prior to further experimentation, is needed. RESULTS: A novel molecular pattern recognition procedure has been developed to diagnose the differentiation state of ES cells. This is based on mRNA transcript levels of genes differentially expressed between ES cells and their differentiating progeny. Large publicly available ES cell data sets from various platforms were used to develop and test the diagnostic model. Signature patterns consisting of five gene expression levels achieved high accuracy at determining the cell state (sensitivity and specificity > 97%). CONCLUSION: The effective ES cell state diagnostic scheme described here can be implemented easily to assist researchers in identifying the differentiation state of their cultures. It also provides a step towards standardization of experiments relying on cells being in the stem cell or differentiating state.published_or_final_versio

    A high-temperature superconducting monolithic microwave integrated Josephson down-converter with high conversion efficiency

    A compact high-Tc superconducting monolithic microwave integrated circuit Josephson down-converter is presented. The circuit consists of a single Josephson junction mixer, a bandpass filter, a lowpass filter, and a resonator for local oscillator fabricated on a single 10 mm × 20 mm chip of YBa2Cu3O7-x film on MgO substrate. The down-converter demonstrates superior performance in terms of conversion efficiency, dynamic range, linearity, and low local oscillator power with stable operation from 20 to 77 K. A maximum conversion gain of -4.7 dB was measured at 20 K and -12.8 dB at 70 K. © 2013 Crown

    Management of Extracranial Carotid Artery Aneurysms:17 Years» Experience

    AbstractObjectives:a retrospective review of seventeen-year (1980–1996) experience of the management of extracranial carotid artery aneurysms.Patients and methods:sixty-six aneurysms of extracranial carotid artery were seen in 63 patients. The diagnosis was confirmed by angiography in 51 patients and duplex ultrasonography in twelve. Twenty-eight (42%) patients had an atherosclerotic aneurysm, twenty-two (33%) had false aneurysms secondary to trauma, nine were congenital and seven were mycotic. All underwent aneurysm resection with saphenous-vein-graft interposition as the most common means of reconstruction.Results:one death occurred due to septicaemia in a diabetic patient with a mycotic aneurysm, giving an operative mortality of 1.5%. One patient had an immediate hemiparesis after carotid artery ligation, and three had a hemiparesis within 48 hours of operation (6.1%). After a change in technique to avoid a residual carotid stump, no further neurological problems were encountered in the following 28 patients.Conclusion:extracranial carotid aneurysms may be successfully managed with resection and reconstruction with autogenous saphenous vein. End-to-side anastomosis avoids a blind-ending stump which may be the source of emboli

    Monolithic high-temperature superconducting heterodyne Josephson frequency down-converter

    A monolithic microwave integrated circuit (MMIC) frequency down-converter based on a compact high-T c superconducting (HTS) device is demonstrated. The on-chip integrated HTS down-converter consists of a 7-9 GHz bandpass filter for RF input, a lowpass filter for intermediate frequency output, and a self-pumped Josephson heterodyne mixer. All the above passive and active components are fabricated on a single 10 mm × 20 mm chip of YBa 2Cu 3O 7-x film on MgO substrate. Characterization of this MMIC HTS down-converter in terms of frequency response, conversion gain, frequency-tuneability, bias dependence, dynamic range, linearity, and intrinsic noise are presented in this paper. © 2012 Crown

    [Fe iii] lines in the planetary nebula NGC 2392

    The Eskimo Nebula (NGC 2392) is a young double-shell planetary nebula (PN). Its intrinsic structure and the responsible shaping mechanism are still not fully understood. We present new optical spectroscopy of NGC 2392 at two different locations to obtain the spectra of the inner and outer shells. Several [Fe iii] lines are clearly detected. We find that these [Fe iii] lines mostly originate from the inner shell. Therefore, we suggest that NGC 2392 might have an intrinsic structure similar to the Ant Nebula Mz 3, which exhibits a number of [Fe iii] lines from the central dense regions. In this scenario, the inner and outer shells correspond to the central emission core and the outer lobes of Mz 3, respectively. © 2012 International Astronomical Union.published_or_final_versio

    Curcumin Enhances Bortezomib Treatment of Myeloma by Inhibiting Heat Shock Protein 90 Expression

    Purpose: To investigate whether curcumin augments bortezomib-induced apoptosis in myeloma cells (MM1.R line), and to explore the molecular mechanism with regard to heat shock protein 90 (HSP90) expression.Methods: MTT cell viability assay was used to assess growth inhibition of MM1.R cells at different concentrations of curcumin alone and also combined with 0.01 mM bortezomib. Annexin V and propidium iodide (PI) labeling were used to detect apoptosis. Caspase 3, caspase 9, NF-κB, and HSP 90 protein expression were measured by Western blotting.Results: Curcumin alone inhibited MM1.R cell growth and increased apoptosis in a concentration dependent manner. When curcumin was combined with low concentration (0.01 mM) bortezomib, both effects(viability inhibition and apoptosis induction increased (p < 0.05), whereas bortezomib alone had no effect (p > 0.05). Western blotting revealed that for curcumin and combined treatments, expression of the apoptotic markers, caspase 3 and caspase 9, increased while expression of NF-κB and HSP 90 decreased (p < 0.05). Again, low concentration bortezomib alone had no effect, whereas the combined treatment showed the largest effect, thus suggesting that the actions of curcumin and bortezomib are synergistic.Conclusion: Curcumin increased MM1.R cell sensitivity to bortezomib, which may be due to suppression of NF-κB and HSP90 activity.Keywords: Curcumin, Bortezomib, Myeloma cells, Cell growth, Apoptosis, Heat shock protein 9

    Localized-Surface-Plasmon Enhanced the 357 nm Forward Emission from ZnMgO Films Capped by Pt Nanoparticles

    The Pt nanoparticles (NPs), which posses the wider tunable localized-surface-plasmon (LSP) energy varying from deep ultraviolet to visible region depending on their morphology, were prepared by annealing Pt thin films with different initial mass-thicknesses. A sixfold enhancement of the 357 nm forward emission of ZnMgO was observed after capping with Pt NPs, which is due to the resonance coupling between the LSP of Pt NPs and the band-gap emission of ZnMgO. The other factors affecting the ultraviolet emission of ZnMgO, such as emission from Pt itself and light multi-scattering at the interface, were also discussed. These results indicate that Pt NPs can be used to enhance the ultraviolet emission through the LSP coupling for various wide band-gap semiconductors

    Growth of GaSb and GaInAsSb layers for thermophotovoltaic cells by liquid phase epitaxy

    GaSb based cells as receivers in thermophotovoltaic system have attracted great interest and been extensively studied in the recent 15 years. Although nowadays the manufacturing technologies have made a great progress, there are still some details need to make a further study. In this paper, undoped and doped GaSb layers were grown on n-GaSb (100) substrates from both Ga-rich and Sb-rich solutions using liquid phase epitaxy (LPE) technique. The nominal segregation coefficients k of intentional doped Zn were 1.4 and 8.8 determined from the two kinds of GaSb epitaxial layers. Additionally, compared with growing from Ga-rich solutions, the growing processes from Sb-rich solutions were much easier to control and the surface morphologies of epitaxial layers were smoother. Furthermore, in order to broaden the absorbing edge, Ga 1-xInxAsySb1-y quaternary alloys were grown on both GaSb and InAs substrates from In-rich solutions, under different temperature respectively
